"Ay" on a long scale

This is "ay" as in "cake" on a long scale. The last note is sustained with vibrato. Seth says to "use your ee lips" and it's likely he's reminding Michael to keep his lips relaxed and not pulled back into a smile. That's a common speech-level technique for handling an "ee" sound without impeding it with accessory facial muscles.
